1921年C.C.Little用Abby Lathrop小鼠近亲培育数个近交系，使编号为57的雄鼠与52号雌鼠交配，培育成C57。其中毛色固定为黑色的培育成C57BL，BL是英文Black（黑色）的缩写。1937年，Little将维持的C57BL父系进行分离，将第6组亚系定名为C57BL/6。美国NIH（National Institutes of Health）引进C57BL/6后进行近交培育，形成的近交系C57BL/6N。适合DIO（Diet-induced obesity mouse，饮食诱导的肥胖）、代谢综合症、脂肪肝、动脉粥样硬化造模或研究。老年时发生迟发性听觉丧失（Age late-onset hearing loss）。对声音和气味很敏感，产生应激反应。拔毛癖或强迫性拔毛行为。酒精性疾病和吗啡成瘾的研究。
